Neutrophils and platelets cooperate in immunosurveillance to safeguard host defence; yet, this interaction can become pathogenic. Here, we link neutrophil substates in thrombus development to functional states. Specifically, we map and isolate thrombus-resident neutrophil subsets and dissect their prothrombotic properties. We trace neutrophil maturation trajectories from the circulation towards and within thrombi. Finally, we use models of altered granulopoiesis to assess how different circulating neutrophil maturation states influence thromboinflammation.
