We study the dynamic interaction of the basolateral polarity regulators LKB1 and Par-1 with phospholipids of the plasma membrane using mammalian cell culture models and Drosophila in vivo experiments. Furthermore, we apply cryo-electron microscopy and molecular simulations to resolve the molecular mechanism of LKB1/Par-1-phospholipid interaction and its impact on protein localization and kinase activation to understand the establishment of apical-basal polarity.
